Environmental DNA meta-barcoding to reveal aquatic insect community in fragmented river

Environmental DNA (eDNA) has been paid attention to as a new biological monitoring method on the aquatic ecosystem. The eDNA meta-barcoding analysis is currently employed to investigate the community composition of various aquatic organisms such as fishes, amphibians, mammals, and insects. This study examined aquatic insect communities in the river fragmented by a dam (Shiokawa River) and non-fragmented river (Kamanashi River) in Yamanashi, Japan, using eDNA meta-barcoding. Water-samplings were performed between Nov 2018 and Nov 2020. This basin was disturbed by heavy typhoon Hagibis in 2019. We collected 500ml of river-water and filtered it by a mixed cellulose membrane filter (pore size 0.2µm). The eDNA was extracted by the phenol-chloroform method or DNeasy Blood and Tissue kit (Qiagen). The Cytochrome Oxidase I region was amplified by PCR and sequenced using Illumina sequencer. The obtained reads were filtered by sequence quality and searched its origin organisms by BLAST.
